张学工,1989年毕业于清华大学自动化系,1994年于清华大学获模式识别与智能系统专业博士学位,现为清华大学自动化系教授、生命学院和医学院兼职教授,清华信息科学与技术国家实验室生物信息学研究部主任,清华大学学术委员会委员,中国人工智能学会生物信息学与人工生命专业委员会主任、中国生物物理学会生物信息学与理论生物物理专业委员会副主任,是国家杰出青年基金获得者、国家级精品课程主讲人、国家973计划首席科学家,主要研究方向是模式识别与生物信息学。
内容提要:
The next-generation sequencing has become a major technology in current biology. Correctly and efficiently reading out the full biological information contained in the massive sequencing data is the foundation of all biological analyses, and is a major task for current bioinformatics. The third-generation single molecular real time (SMRT) sequencing technology not only provides an opportunity for obtaining very long reads, but the real-time nature of the sequencing reaction leaves signatures of DNA modifications in the obtained sequencing signals. We investigated the characteristics of these signals and developed a flexible hierarchical model for detecting DNA modifications from SMRT sequencing data. The application on bacteria genome sequencing data indicated the existence of epigenetic heterogeneity in a cell population, and we further developed an empirical Bayes mixture model to quantitatively estimate this heterogeneity of DNA modification. The method was validated on both simulation and bacteria DNA modification data.
